The 30-year Exeter star won six England caps and last appeared for the country in 2018.
Leicester tighthead Will Hurd, Sale winger Arron Reed and Edinburgh back Harry Paterson are the other three new additions in Gregor Townsend's squad.
Northampton back Rory Hutchinson will be seeking to add to his eight caps as he returns for the first time since appearing in two Tests against Argentina in 2022. Gloucester stand-off Adam Hastings is back in after missing the World Cup last year through injury.
Former Wallaby Jack Dempsey and Glasgow teammates Rory Darge and Kyle Steyn made the selection despite recent injury lay-offs.
Darge is unlikely to be fit until midway through the tournament, which starts for Scotland with a trip to Wales on February 3.
The most notable absentees from Townsend's squad are 2021 British and Irish Lions trio Hamish Watson, Rory Sutherland and Chris Harris.
Dave Cherry, who departed the World Cup squad in September after falling down stairs at the team hotel in Nice, has also been omitted.
Meanwhile, new Italy coach Gonzalo Quesada on Tuesday named five new faces in his preliminary 34-man squad.
Among the new names in the squad is South-African born Ross Vintcent, the young Exeter flanker making the step up from the under-20s. The other four debutants are also among the forwards.
There are also a host of familiar faces from the team which under Quesada's predecessor Kieran Crowley made a mark in international rugby with an ambitious playing style.
Those include Toulouse full-back Ange Capuozzo, the undoubted star of the team, Montpellier fly-half Paolo Garbisi and Gloucester scrum-half Stephen Varney.
Quesada will cut back the squad following a training camp in Verona next week, ready for Italy's Six Nations opener at the Stadio Olimpico.
As for Wales and Warren Gatland, Exeter lock Dafydd Jenkins was appointed Wales captain as Jac Morgan, who skippered them at the World Cup, is likely to miss the Six Nations after knee surgery.
Gatland named five uncapped players in Cardiff quartet Cameron Winnett, Evan Lloyd, Alex Mann and Mackenzie Martin, plus Bath prop Archie Griffin.
Flyer Louis Rees-Zammit confirmed he will not take part as he attempts to make the NFL.
